The SIF protein, containing Dbl Homology (DH) domain, is homologous to GDP-GTP exchange factors, especially to Tiam-1, which convert Rho-like G proteins from a GDP-bound inactive state to a GTP-bound active state. SIF protein also possesses Pleckstrin Homology (PH) and PDZ domains, both of which are found in various signaling molecules. SIF protein was found adjacent to the plasma membrane of synaptic terminals. Expression of a truncated SIF protein induced membrane ruffling with altered actin localization in human KB cells. Thus, SIF protein may regulate synaptic differentiation through the organization of the actin cytoskeleton by activating Rho-like G proteins.

Fujita Cranial sensory neurons mediating special senses such as hearing and taste project to ganglion-specific targets both in the periphery and in the brain. Our hamster monoclonal antibody 802Cll intensely stained neurons and their processes of the VIIIth cranial ganglion of the mouse embryo, but not those of the Vth or spinal dorsal root ganglia. Biochemical analyses of the antigen from adult mouse brain showed the antigen to be a glycosylated intrinsic membrane protein of ca.100 KDa. The protein was purified, and based on partial amino acid sequences determined of proteolytic fragments, the entire cDNA was cloned. The deduced amino acid sequence had a high homology (73.5% identity) to chicken SC1 protein, a cell-cell adhesion molecule belonging to the immunoglobulin superfamily with a proposed role in neurite growth of spinal motor neurons. Mouse SC1 may be involved also in the growth of specific cranial sensory nerve fibers and neuronal network formation.

Expressin of PLP and PDGFa-R gene were examined in mouse brain during development by ISH. In contrast to PLP-mRNA, PDGF-(r-R-mRNA was detected in ganglionic eminence of brain at El 1, spinal cord at El2 and nervous system at El4 and thereafter. These results suggest many origins for oligodendrocytes.
