Sung-Sik Han 1, In-Cheon Hwang 2 1Dept. of Agricultural Biology, Korea University, 136-701 Seoul, Korea, and 2Kyungju Research Institute, Kyungnong Corporation, 780-110 Kyungju, Korea Structure of marginal band of microtubules (MB) and their assembly process in the immunocyte of Blattella germanica have been investigated via TEM, lytic medium treatment and immuno-fluorescence. Of particular interest is the lytic medium treatment that produces the remarkable result for features of MB. Formation of MB begins with two MBs originated in MTOC. Each MB grows in the opposite direction drawing a more or less circle in outline. Their growth continues until each MB comes back to the starting point at MTOC, hence, establishing 2 circular MBs upon completion. Comparison of the number of microtubules at different stages also supports this scheme. Microtubules in a band has been increased approximately double in number beyond the overlapping point.
