Objective: To evaluate relationship of endothelial dysfunction with blood pressure (BP) daily profile and type of left ventricle (LV) remodeling in essential arterial hypertension (AH). We examined 124 patients with AH with LV-function, vasomotor endothelial function, daily BP monitoring assesments. Results: Diameter of brachial artery was the greatest in patients with profile type night-peaker, and was 4.25(3.80;5.00), non-diper e 4.20 (3.70;4.60), dipper e 4.10(3.50;4.65) mm Hg, reduced and most patients over-dipper, while the velocity of the blood was reduced in night-peaker patients e 0.60(0.53;0.84) m/s. Vasodilatatory reaction during decompression in night-peaker patients was 5.47(3.04,11.72)%, p<0.00014, in non-dipper patients was 11.63(7.76;18.92)%, dipper e 8.94(7.04;15.46)%, over-dipper e 7.24(5.82;13.32)%. Vasodilatatory response to nitroglycerin was preserved in most patients with non-dipper - 16.6(10.52;25.00)%, night-peaker e 13.67(8.01;20.24)%, over-dipper e 11.72(7.54;17.08)%, and decreased in dipper patients e 9.97(7.04;15.46)%. There is inverse correlation between endothelial dysfunction and concentric hypertrophy (r¼-0,32;p¼0.001), type of BP non-dipper (r¼-0.27;p¼0.009), degree of night reduction in diastolic BP (r¼-0.25;p¼0.014), systolic index (r¼-0.25;p¼0.016), dilatation of brachial artery with nitroglycerin (r¼0.24;p¼0.017),, the type of dipper (r¼-0.22;p¼0.032), degree of night reduction in systolic BP (r¼-0.22;p¼0.030), speed E (r¼-0.21, p¼0.037). Direct correlation was between the degree of endothelial dysfunction and degree of diastolic dysfunction (r¼0.37;p¼0.00038), concentric and excentric hypertrophy (r¼0.25, 0.23; p¼0.015, p¼0.02), restrictive type of diastolic dysfunction (r¼0.25;p¼0.026), normal LV geometry (r¼0.21;p¼0.036), type of night-peaker (r¼0.23;p¼0.026), LV systolic dysfunction (r¼0.21;p¼0.036). Conclusion: In AH endothelial dysfunction is factor of correlation with LVremodeling by concentric type, and with change of BP circadian rhythm as night-peaker.

Although the pathophysiology of atherosclerosis is not completely understood, inflammation is regarded to play a major role in its development. Recently we reported that induction of peroxisome proliferator-activated receptor-GAMMA (PPARGAMMA) selectively inhibits vascular cell adhesion molecule-1 (VCAM-1) but not intercellular cell adhesion molecule-1 (ICAM-1) in tumor necrosis factor (TNF)-ALPHA-activated human umbilical vein endothelial cells (HUVEC). In this study, we investigated whether genipin, a main constituent of gardenia, inhibits expression of cellular adhesion molecules, which is relevant to inflammation. Pretreatment with genipin reduced reactive oxygen species (ROS) production and expression of VCAM-1, but not ICAM1 in TNF-ALPHA-activated HUVEC. Genipin dose- and time-dependently increased PPAR-GAMMA expression and inhibited TNF-ALPHA-induced phosphorylation of Akt and PKC with different degrees. Finally, genipin prevented TNF-ALPHA-induced adhesion of U937 monocytic cells to HUVEC. Taken together, these results indicate that upregualtion of PPARGAMMA by genipin selectively inhibits TNF-ALPHA-induced expression of VCAM-1, in which regulation of Akt and/or PKC play a key role. We concluded that genipin can be used for the treatment of cardiovascular disorders such as atherosclerosis. EAS-0302. High IgE antibody level in the serum of patients with unstable angina as compared to healthy individuals as well as presence of IgE and its receptor FcεR in human atheromatous plaques suggest potential involvement of IgE in atherosclerosis pathogenesis. Therefore the purpose of the study was to demonstrate the presence of FcεR mRNA on endothelial cells (HAEC). An additional purpose of the work
