We now hemonstrate the exist&e of a novel form of cellular cytotoxicity that is plasma and cell activation dependent. It is mediated both bv human peripheral blood hononuclear cells (PBM) and “by the transformed human monocytic cell line TIH-1. PDM and THP-I. when stimulated by LPS and in the presence of plasma, are cytotoxic to murine WEHI 164 cells in a 6 hr 51 Cr-release assay. Neither fresh nor heat-inactivated strum substitutes for ~plasma. We propose the designation of Plasma Dependent Cellular Cytotoxicity (PDCC) and suggest that this mu represent a novel effector mechanism in the host immune anb inflammatory response to transformed and other divergent cells.

Schl. of Med., Phila., PA 'Biosearch Laboratories, Inc., Phila., PA Five males, 61, with severely p otodamaged skin received 0.057 tretinoin cream (Retin-A & , Ortho Pharmaceuticals) twice daily for one month to the skin of the dorsal forearm. The following procedures were done before and after treatment: (1) 3 mm punch biopsies for histochrmistrv, (2) sticky slides for degree of melanization of corneocytes, (3) detergent scrubs for corneacyte sizing and morpholagy, and (4) Silflo replicas for surface topography. The epidermal histologic abnormalities were largelv corrected by one month of treatment. Atypia and dysplasia were eliminated accompanied by acanthanosis, restoration of polarity and a thinned horny layer. Dermal changes were insignificant. Surface glyphic patterns reappeared. 'melanin granules within corneocytes were greatly decreased, associated with modest bleaching of

Although some Investigators have already reported the detection of Mycobacterium leprae using the polymerase chain reaction (PCR), PCR is still not employed as a screening test in developing countries where such diagnostic systems are most needed. The purpose of this study was to highlight the problems involved in PCR detection of M. leprae and to suggest possible improvements in the practical use of PCR as a screening test even in developing countries. Our findings demonstrated the necessity for selectjon of highly specific primers, the most suitable samples for the screening test. It is concluded that a complete diagnostic system for leprosy using PCK can be established wlthout the need for radioisotopes, electrophores?s or membrane hybridization, which would enable a screening test to be done by PCR even in developing countries.
