Toll-like receptors (TLRs) function by activating MyD88-dependent and -independent signaling pathways. In this study, mice were generated with a genetic deficiency in a Toll/IL-1 receptor (TIR) domain–containing adapter protein (TRIF) and were analyzed for their response to Toll stimuli. Notably, TRIF-deficient mice were defective in both TLR3- and TLR4-mediated expression of IFN-β. In addition, inflammatory cytokine production triggered by TLR-4 ligand (but not other TLR ligands) was severely impaired in cells derived from TRIF-deficient mice. Only mice deficient in both MyD88 and TRIF had complete loss of NF-κβ activation in response to TLR-4 stimulation. Collectively, these findings indicate a central role for TRIF as an adapter molecule in stimulating MyD88-independent TLR signaling pathways.
