EFFECT OF POPULATION DENSITY ON ELICITATION OF CONTACT HYPERSENSITIVITY. -oil. T. Tsucl&&R& win* J Kovd ’ The Life Science Laboratories, Shiseido, Tokyo, Japan, ‘Department of Dermatology, Cornell University, New York, USA To reveal effects of sqcial stress on immune reactions in the skin allergic contact hypersensitivity (CH) was elicited in mice housed at various population density. Female C57BU6CRj mice were housed at the density of 5 per cage and sensitized by application of TNCB on the dorsal skin. After 4 days the density was changed to 1,5, or 10 per cage. On the next day mice were challenged at ears by painting of TNCB. Ear swelling was measured and the blood was collected to assay for corticosterone after 24 hours. Corticosterone increased as the density increased, demonstrating that high density is stressful for animals. The elicitation of the CH was less in mice housed at high density (i/cage: O.l78mm, 5: 0.134, 10: 0.088). These results suggest the regulation of skin immune reactions by social stress.

In this study, we investigated what kind of cytokines are produced by regional lymph node stimulated with a bacterial superantigen cells or a specific hapten in murine contact hypersensitiVity(CH). BALB/c mice were sensitized with dinitrofluorobenzene and the r.egional lymph node cells were cultured with a specific hapten. Several cytokines in the culture supernatant were measured. Interferon-r (IFN-y ) was mainly produced by the lymph node cells. Staphylococcal enterotoxin B (SEB) may affect CH responses as a immunomodulator because normal lymph node cells stimulated with SEB produced some cytokines including IFN-7 .
