Sci., Okazaki 4443 YUIUE NAKAMOTO’ , TSUYOSHI MIYAKAWA2, TAKESHI YAG13, HIROAKI NIK12, MITSUNOBU YOSHII’ Fyn tyrosin’e kinase deficient mice progress audiogenic seizure susceptibility and fearfulness (Miyakawa et al., 1995; 1996). The density of central- type benzodiazepine receptors (CBR) decreases in the brain of Fyn-deficient mice whereas that of peripheral-type benzodiazepine receptors (PBR) shows no significant difference between Fyn-deficient and control mice (Nakamoto et al., 1996). In the present study, we have further investigated distributions of the CBR and the PBR by in vitro quantitative autoradiography using [3H]flunitrazepam and [3H]PK11195 as ligands for CBR and PBR, respectively.In Fyn-deficient mice, [3H]flunitrazepam binding was decreased widely, particularly in front-parietal cortex, hippocampus, globus pallidus and amygdala, whereas rH]PK11195 binding showed no significant alterations. The results suggests that seizure susceptibility and fearfulness in Fyn-deficient mice may result from dysfunction of the GABAergic system in these brain regions.
