PATENT ABSTRACTS
A method is provided for restoring some or all of the activity of gamma-interferon which has been in contact with an acidic solution comprising the steps of: (a) placing the gamma-interferon in a solution which has a pH between about 5.5 and 9.5; and (b) incubating the solution at a temperature of between about 2 degrees C. and 8 degrees C. for a period of at least 24 hours. The method allows antibody affinity chromatography employing acid elution to be used to purify gammainterferon, in that, the activity of the acid-eluted gamma-interferon can be essentially completely restored using the reactivation process.
4499184 ANALYSIS FOR TOTAL CHOLESTEROL USING LECITHIN:CHOLESTEROL ACYL TRANSFERASE (LCAT) Charles T Goodhue assigned to Eastman Kodak Company Methods, compositions and elements for the analysis of total cholesterol in aqueous fluids containing cholesterol and cholesterol esters are disclosed. Cholesterol esters in the fluid samples are first converted to cholesterol by: (a) contacting a sample of the fluid with sufficient amounts of lysolecithin and an enzyme exhibiting lecithin:cholesterol acyl transferase (LCAT) activity to convert substantially all the cholesterol esters to cholesterol in the sample, and (b) removing cholesterol from the aqueous fluid other than by conversion to cholesterol esters. The total cholesterol is quantitatively determined by measuring the amount of the removed cholesterol. For example, the amount is measured gravimetrically or by converting the cholesterol to a detectable product.
4499185 TEST FOR ESTERASE ACTIVITY IN A LIQUID SAMPLE A Christophe Skjold, Lonnie R Stover, Robert W Trimmer assigned to Miles Laboratories Inc A composition, test device and method for determining the presence of teukocytes, esterase and/or esterase activity in a test sample is disclosed. The composition comprises an ester which, when cleaved by esterolytic activity, produces a detectable response such as color formation. In addition the composition comprises the compound 3-quinuclidinol. The device is prepared by incorporating a carrier matrix, such as paper, with the composition. The method comprises contacting a test sample with the device and observing a detectable response.

4499186 DIAGNOSING AUTOIMMUNE R H E U M A T O I D A R T H R I T I S BY MEASURING PROTEOLYTIC ACTIVITY OF ALPHA2MACROGLOBULIN Marius C Teodorescu, Alexander M Gaspar, Gregory T Spear, Doina Ganea, John Skosey assigned to University of Illinois Board of Trustees A method for diagnosis of rheumatoid arthritis and related autoimmune diseases comprises blocking calcium ions contained in a blood sample, effecting hydrolysis of a selected substrate in the presence of alpha2-macroglobulin ( alpha2M) from the blood sample, and determining the extent of hydrolysis of the substrate. Preferably, alpha2M in plasma is incubated with a hydrolyzable chromogenic substrate and the liberated chromogen is determined spectrophotometrically. A diagnostic kit is also provided.
4499187 METHOD FOR EVALUATING MUTAGENICITY Gary Blackburn, Carl Mackerer, Ceinwen A Schreiner assigned to Mobil Oil Corporation A method is provided for rapidly and reliably determining the potential carcinogenic activity of hydrocarbon mixtures which is especially useful for those of petroleum origin. A sample of the mixture is extracted with dimethylsulfoxide (DMSO). An inoculum of Salmonella typhimurium tester strain T98 is incubated in the presence of an aliquot of the extract together with an optimal amount of induced rat liver homogenate $9. The excessive production of revertant colonies is a measure of the mutagenic activity of the oil, and this measure is shown to correlate with dermal carcinogenic activity.
