LABELS

Lyle J Arnold, Alexander Waldrop, Philip Hammond assigned to Gen-Probe Incorporated A method of preparing a labelled specific binding partner, such as a biological probe in the form of an antibody or oligonucleotide probe, using a protected label (the corresponding unprotected label being susceptible to inactivation, such as by hydrolysis to yield a nonchemiluminescentform of the label). The specific binding partner is linked to the label, and an adduct of the label is prepared using a protective adduct former which produces a protected label which is less susceptible to inactivation. Particularly preferred labels are the acridiniums and acridans, most preferably the former having the general structure: See Patent f o r Chemical Structure wherein the phenyl rings are optionally additionally substituted, RI is preferably an alkyl, and R5 is an optionally substituted hydrocarbon, most preferably a phenyl moiety which is either linked to the specific binding partner, or capable of being linked thereto. Formation of the protected label is preferably an equilibrium reaction which is readily reversible, such as by dilution or oxidation of the protective adduct former. Labels useful in such a method are also disclosed.

4950647 T CELL IMMUNOPOTENTIATOR Roland K Robins, Brahma S Sharma, Ganapathi R Revankar assigned to Nucleic Acid Research Institute See Patent f o r Chemical Structure 6-Amino-1-

(2-deoxyalphaD-erythro pentofuranosyl)imidazo (4,5-C)pyridin-4-one (also know as alpha-2'-deoxy 3-deazaguanosine) is non mitogenic to human peripheral blood lymphocytes; however, it displays a potent immunoenhancing activity on human T cells.

This compound causes a marked increase in both phytohemagglutinin and Concanavalin A induced proliferation of T cells. When added during allogeneic mixed lymphocyte response, it also potentiates the proliferation ofalloreative T cells. The compound shows no potentiating effect on B lymphocytes activated either with Staphylococcus aureus cowan or Pokeweed mitogen, suggesting that it is mainly a T cell function immunopotentiator. The compound overcomes induced immunosuppression and is able to restore depressed proliferative responses ofT cells. Further it potentiates the generation of antigen-specific primary cytotoxic T lymphocytes.

49~7~ RECOMBINANT DIPHTHERIA VACCINES Lawrence Greenfield, Anne W Emerick, Walter J Laird assigned to Cetus Corporation Recombinant diphtheria toxin A fragment muteins which are enzymatically inactive but immunologically crossreactive with diphtheria toxin are disclosed. Intermediates and methods for preparing such proteins using recombinant techniques are also described.

4950741 ANTIBODY AGAINST RHEUMATOID ARTHRITIS SPECIFIC PROTEIN Naoki Yamanaka, Makoto Yoshida, Nagoya, Japan assigned to Asahi Medical Co Ltd; Medecs Co Ltd A substantially pure rheumatoid arthritis specific protein (RASP) and an antibody against the rheumatoid arthritis specific protein (antiRASP antibody) are disclosed. The RASP is found specifically in the serum or plasma of a patient suffering from rheumatoid arthritis, and may be detected using an anti-RASP antibody easily and effectively. Therefore, the anti-RASP antibody of the present inventionis useful for the diagnosis of rheumatoid arthritis by the criterion of the presence of RASP.
