You Kwan-sa - Duke University Salicylate hydroxylase isolated from Pseudomonas bacteria can be used to determine the level of salicylate in a body fluid by reacting a sample of the fluid with the enzyme and monitoring the conversion of salicylate to catechol. A method of purifying the enzyme from crude bacterial extract using a salicylate affinity column is also disclosed.

The method comprises contacting the test sample with an indicator reagent composition which interacts with the peroxidatively active substance to produce a measurable or detecable color change. The indicator reagent composition comprises a redox indicator dye, a hydropero~de, a ferric ion and a phosphorous compound having the formula (*See Patent for Chemical Structure*) wherein R1 or R2 are certain substituted or an unsubsfituted aromatic moieties or certain polyhydric compounds, and wherein X is O, S or NH. The assay can be performed in wet phase or the composition can be incorporated into a carrier matrix. The method provides a more accurate and trustworthy assay of a test sample for a peroxidatively active substance and is especially useful in the assay of urine for occult blood.

There is disclosed a process for the fermentation by yeast of sugar to alcohol wherein a portion of the nutriem phosphate is provided by one or more condensed phosphates. It has been found that when condensed phosphates are employed as the nutrient source the fermentation rate increases. In addition, the rate increase is only observed when an increase in macronutrient elements is provided in conjunction with the condensed phosphates to compensate for the elements sequestered by the condensed phosphates.
