In v i v o screening of potential immunosuppressants by s k i n grafting or heterotopic heart transplantation is slow, labor-intensive, and costly. We t h e r e f o r e d e v e l o p e d a s e n s i t i v e t e s t b a s e d on t h e r e j e c t i o n of t h e a l l o g e n e i c l e u k e m i a L I 2 1 0 (H-2 d) by C 5 7 B L / 6 (H-2 b) mice. R e j e c t i o n of t h e L l 2 1 0 c e l l s w a s d e t e r m i n e d 7 d a y s a f t e r an i n t r a v e n o u s c h a l l e n g e of I0' c e l l s by r e m o v i n g t h e spleens, preparing a single-cell suspension, a n d a n a l y z i n g t h e c e l l s for t h e p r e s e n c e of H 2 K d - p o s i t i v e (LI210) c e l l s . E n u m e r a t i o n of t h e a l l o g e n e i c c e l l s w a s performed by flow cytometry using a FITC-labelled anti-H2Kdmonoclonal antibody or its i s o t y p e c o n t r o l . Non-immunosuppressed mice rejected the LI210 cells and survived indefinitely. S p l e e n s f r o m t h e s e m i c e w e r e n e a r l y f r e e of H 2 K d - p o s i t i v e c e l l s b y d a y 7 (0.34 + 0 . 1 4 % ) . In c o n t r a s t , m i c e t r e a t e d w i t h i0 m g / k g / d a y of cyclosporine (CsA) had--ll.l + 1.5% H 2 K d - p o s i t i v e c e l l s on d a y 7. M i c e t r e a t e d in t h i s m a n n e r h a d a m e a n s u r v i v a l t i m e of 9+1 day. M i c e t r e a t e d w i t h 50 m g / k g / d a y of C s A h a d 17.8 + 3.2% H 2 K d - p o s i t i v e c e l l s . We o b s e r v e d a d i r e c t c o r r e l a t i o n b e t w e e n t r o u g h l e v e l s of C s A in t h e b l o o d a n d t h e p e r c e n t a g e of H 2 K d - p o s i t i v e c e l l s (r=0. 958).

Endometriosis (EM) is an i n t r a p e r i t o n e a l i n f l a m m a t o r y d i s e a s e a s s o c i a t e d w i t h c h r o n i c pain and i n f e r t i l i t y . The d i s e a s e is c h a r a c t e r i z e d by the i m p l a n t a t i o n of e n d o m e t r i a l t i s s u e in sites o u t s i d e the u t e r u s . It has been s u g g e s t e d that EM may be l i n k e d to a l t e r e d i m m u n e f u n c t i o n , but the i n t r a p e r i t o n e a l immune r e a c t i v i t y in these p a t i e n t s is not clear. T h i s study e v a l u a t e d s u r f a c e a n t i g e n e x p r e s s i o n on p e r i t o n e a l l y m p h o c y t e s f r o m w o m e n w i t h m i l d EM r e l a t i v e to c o n t r o l s free of this d i s e a s e . F l o w c y t o m e t r i c a n a l y s i s of a n t i g e n e x p r e s s i o n r e v e a l e d a C D 4 : C D 8 ratio of 0.6 for p e r i t o n e a l T cells from b o t h c o n t r o l and EM p a t i e n t s . R e l a t i v e to c o n t r o l s , h o w e v e r , T cells from EM p a t i e n t s e x h i b i t e d s i g n i f i c a n t l y e l e v a t e d H L A DR e x p r e s s i o n and also s h o w e d i n c r e a s e d e x p r e s s i o n of i n t e r l e u k i n 2 r e c e p t o r ( C D 2 5 ; T A C ) . In c o n t r a s t , EM p a t i e n t s had s i g n i f i c a n t l y r e d u c e d levels of p e r i t o n e a l l y m p h o c y t e s u b s e t s c o - e x p r e s s i n g C D 2 / C D 2 9 r e l a t i v e to c o n t r o l s . Cell cycle a n a l y s i s d e m o n s t r a t e d a t w o - f o l d i n c r e a s e in the p e r c e n t a g e s of cells from EM p a t i e n t s in S phase, c o m p a r e d to n o r m a l c o n t r o l w o m e n ° T a k e n t o g e t h e r , these data s u g g e s t that the p e r i t o n e a l l y m p h o c y t e s of w o m e n with e n d o m e t r i o s i s c o n t a i n p o p u l a t i o n s of a c t i v a t e d T cells° The a l t e r e d f u n c t i o n a l a c t i v i t i e s of these l y m p h o c y t e s u b p o p u l a t i o n s may c o n t r i b u t e s i g n i f i c a n t l y to the d i s e a s e p r o c e s s .

H o w e v e r , it has not yet b e e n f u l l y s t u d i e d . We e x a m i n e d the h i s t o p a t h o l o g i c a l and immunoh i s t o c h e m i c a l a s p e c t s of c a r d i a c b i o p s y t a k e n f r o m 20 p e d i a t r i c p a t i e n t s w i t h AVB. P a t i e n t s w i t h c a r d i o m y o p a t h y , s p e c i f i c h e a r t m u s c l e d i s e a s e or c o n g e n i t a l heart disease were eliminated. Advanced pathology including myocyte degener a t i o n , m u s c l e f i b e r d i s a r r a n g e m e n t and f i b r o s i s was o b s e r v e d in 7 / 2 0 ( 3 5 % ) cases, some of w h i c h d e m o n s t r a t e d m o n o n u c l e a r c e l l i n f i l t r a t i o n . I m m u n o h i s t o c h e m i c a l s t u d y s h o w e d T l y m p h o c y t e i n f i l t r a t i o n in the i n t e r s t i t i u m . I m m u n o r e a c t i v i t y for c y t o k i n e (IL-18) w a s not d e t e c t e d in the m y o c a r d i u m . T h e s e r e s u l t s s u g g e s t t h a t s o m e c a s e s of i d i o p a t h i c A V B m a y be r e l a t e d to m y o c a r d i t i s in c h r o n i c or i n a c t i v e s t a t e ( p o s t m y o c a r d i t i c s t a t e ) .
