!J.aM!L we have previously reported that intracerebroventricular (i.c.v.) injections of senktide, a tachykinin NK-3 agonist, showed an antidiuretic action in water-loaded rats (4.5 % body weight)(l). In the present study, we examined the site of action of senktide in the brain on antidiuretic actions in conscious rats. The microinjection of a tachykinin NK-3 agonist, senktide (0.2 - 0.002 nmol) into the paraventricular nucleus (PVN) inhibited urine output in a dose-dependent manner. But substance P (0.2 nmol), tachykinin NK-1 agonist did not inhibit urine output. The antidiuretic effect of senktide was inhibited by pretreatment with a non-peptide vasopressin V2 antagonist (i.v.), OPC-31260. These results suggest that the site of action of the NK-3 agonists for its antidiuretic effect may be PVN of the hypothalamus and that its action may be due to stimulation of (I) Neurosci.Lett.159: 187-190 (1993). vasopressin secretion from the pituitary gland.

Acetylcholine (ACh) and cholecystokinin (CCK) in mammalian frontal cortex have important physiological roles on memory and learning. Systemic treatment with muscarinic antagonist scopolamine and cerebral activator indeloxazine increased ACh output in rat frontal cortex studied with an in vivo microdialysis method. To detect CCK immunoreactivity (CCKLI) in the dialysate of rat frontal cortex, we a highly sensitive enzyme immunoassay using C terminal-directed developed gastrin/CCK antiserum. This assay method was detectable as little as 0.2 Pg CCKa/tube. These results that this method is applicable suggest assay simultaneously to determine both ACh and CCKLI output in the dialysate.
