Ghayour-Mobarhan) Introduction: A randomized controlled clinical-trial in obese subjects was performed to examine the effectiveness of auricular acupuncture on body weight loss and its impact on lipid profile, immunologic and inflammatory markers. Materials and methods: Participants (n = 204), were randomized to therapeutic acupuncture and control groups. Subjects received authentic (cases) or sham (controls) acupuncture for 6-weeks (firstperiod) in combination with a low-calorie diet. In the next 6-weeks (second-period), the low-calorie diet was used on its own. Subjects were assessed at the beginning, 6 and 12 weeks later. In addition to anthropometric and lipid parameters, serum anti-heat shock protein (Hsp)-27, 60, 65, 70 and high sensitive C-reactive protein (hs-CRP) levels were assessed. Results: In the first-period, anthropometric parameters, and hsCRP changed significantly in both groups, while changes in anti-Hsp antibodies were observed only in cases. In the second-period which shows the sustainable effects of acupuncture, changes in anthropometric parameters were more prominent in controls. While significant reductions in group receiving authentic acupuncture were maintained for anti-Hsp antibodies. A comparison between the first and second-period in both groups showed that the changes for most of the parameters were more significant in the first-period. Comparison between cases and controls showed that authentic acupuncture was more effective in reducing the levels of anthropometric factors and anti-Hsp antibodies but not hs-CRP. Conclusion: Auricular acupuncture in combination with diet restriction was effective for weight loss, and dyslipidemia. Moreover, it was found that it has immunomodulatory but not anti-inflammatory effects on immune system by regulation of the levels of anti-Hsp antibodies. E Poster – [A-10-885-1] Higher levels of anti-Hsp27 antibody levels in patients with pemphigus vulgaris compare with healthy subjects Khojasteh-Taheri Roshanak, Yazdan Panah Mohammad Javad, Tavallaie Shima, Rahsepar Sara, Momenzadeh Akram, Ghayour-Mobarhan Majid, Rahsepar Amir Ali, Barzegar Amini Maral Biochemistry & Nutrition Research Center, Mashhad University of Medical Sciences, Mashhad, Iran E-mail addresses: [email protected] (K.T. However, several auto-antibodies have been found in pathogenesis of pemphigus, but some of are still unknown. Antibody to heat shock protein (Hsp)-27 have been reported in several autoimmune diseases including atherosclerosis. Thus, we aimed to evaluate that anti-Hsp27 antibody levels differ between patients with pemphigus vulgaris and healthy control subjects. Material and methods: Twenty-two patients with pemphigus vulgaris were selected and antibody levels against Hsp27 were compared with 22 healthy subjects. Antibody levels were determined by using an in-house enzyme-linked immunosorbent assay (ELISA).

Results: The analysis showed that in patients with pemphigus vulgaris the median and interquartile levels of anti-Hsp27 antibody levels were 0.41 (0.31-0.50), while in healthy subjects the median levels were 0.16 (0.04-0.28). The anti-Hsp27 titers significantly differed between 2 groups (P < 0.001). The antibody levels did not differ statistically between males and females. Conclusion: Our results indicate that anti-Hsp27 antibody is involved in pathogenesis of autoimmune blistering disease of pemphigus vulgaris and can be used in as a potential target for the treatment of pemphigus. Furthure studies are required in order to illustrate that anti-Hsp27 antibody is associated with severity of pemphigus or not. Also, larger sample size is desirable. E Poster – [A-10-904-1] Chromomycin A3 correlate with aniline blue; sperm staining methods to evaluation of protamine deficiency Zandemami Mahdi, Qujeq Durdi, Akhoondi Mohammad Mahdi, Kamali Kourosh, Sadeghi Mohammad Reza Avicanna Research Institute E-mail addresses: [email protected] (Z. Mahdi), [email protected] (Q. Durdi), [email protected] (A.M. Mahdi), [email protected] (K. Kourosh), [email protected] (S.M. Reza) Introduction: Sperm nuclear matrix has a highly organized and packaged structure which is made by protamines. Sperm chromatin packaging anomalies are associated with poor fertility outcomes. Furthermore, human sperm protamine deficiency significantly correlates with diminished sperm functional ability. It has been demonstrated that chromomycin A3 (CMA3) staining correlates with protamine deficiency and in this study it was decided to evaluate the correlation between CMA3 and Aniline Blue (AB) staining to apply one instead of each other. Material and methods: Semen samples from 150 oligoasthenoteratospermic (OAT, case group) and 180 normospermic men (control group) were collected from infertile couples attending to Avicenna Infertility Clinic, affiliated to Avicenna Research Institute (ARI). Semen analysis was performed according to WHO criteria. PBSwashed-sperm slides were prepared in duplicate and after fixation in ethanol:acetone (1:1) were stained with CMA3 and AB. Stained sperms were counted using fluorescent microscopy (for CMA3 staining) and light microscopy (for AB staining). Results: A significant positive correlation was observed between CMA3 and AB (R = 0.75, P < 0.001). The mean of CMA3 and AB results as protamine deficiency marker were significantly higher in OAT group than normospermic group (CMA3: 43.67 ± 17.20% vs. 24.71 ± 13.08%, AB: 39.42 ± 19.11% vs. 20.87 ± 12.59%, P < 0.001). Also result of CMA3 and AB stained sperm had a negative correlation with concentration and motility of sperms (CMA3: R = − 0.426 and −0.232. AB: R = −0.370 and −0.163 respectively, P < 0.001) but did not correlate with morphology of sperm. Conclusion: According to correlation between CMA3 and AB, despite of CMA3, AB can also be applied to protamine level evaluation.
