[email protected] (A. Gokce) BMP-7 and TGF-B1 both stimulate chondrocyte proliferation (CP). Hydrogel based controlled delivery systems for growth factors were designed to form ex vivo a clinically functional tissue for biologicals and mechanical support for chondrocytes during growth and repairing of deficient cartilage areas. The signalling timing of BMP-7 and TGF-B1 may affect CP and eventually a synergism may be seen conspicuously in consecutive controlled delivery. Four groups of cultured chondrocytes were seeded on a novel designed chitosan based hydrogel. The hydrogel was loaded with either BMP7or TGF-B1 or their combination; the control group contained no BMP7/TGF-B1. Hydrogel structure was analyzed by SEM. The release kinetics of Growth Factors (GFs) was determined with ELISA. Chondrocyte viability and toxicity were tested with MTS ELISA Chitosan based hydrogel allowed controlled release of GFs at different time intervals for BMP7/ TGF-B1. A double peak concentration gradient was seen in the group loaded with both GFs. Also in this group we detected significantly higher chondrocyte counts. Canonical regression was used for statistical analysis. Hydrogel systems may be adjusted to release GFs with different molecular weights consecutively and showed a beneficial effect on the growing cell number. doi:10.1016/j.copbio.2011.05.344

[email protected] (M. Yazici) Satureja hortensis which is an annual herb used as nostrum in Eastern Anatolia region of Turkey for the treatment of different infectious diseases and disorders. The aim of the present study was to develop an antifungal activity of liposomal formulation of essential oil of S. hortensis incorporated into the ointment. The liposome formulations were designed using thin film technique and then liposomes were properly incorporated into the ointment. The main active ingredient (Carvacrol) amount was determined by HPLC
technique. In additionally, the chemical composition of essential oil obtained from S. hortensis was determined by GC and GC–MS analysis. The liposomal essential oil of S. hortensis was tested against to Candida albicans with disc diffusion assay and micro-well dilution assay. The antifungal tests results showed that the essential oil of S. hortensis incorporated into the ointment and liposomal essential oil formulation have potential antifungal activity against Candida albicans. The results lead us know that the use of liposomal formulation improves the cellular transport and liposomal essential oil dramatically increased antifungal activity. doi:10.1016/j.copbio.2011.05.345

[email protected] (S. Cengiz) Most algae species synthesize secondary metabolites which might have industrial importance in pharmacy, cosmetics or medicine. The present study evaluated the inhibitory effect of the main secondary metabolite of Caulerpa species, caulerpenyne (CYN), on xanthine oxidase (XOD) enzyme activity, which is considered as a target for the treatment of gout. XOD activity was measured in phosphate buffer solutions (0.05 M, pH 9.00) by measuring the increase in absorbance at 292 nm using xanthine as substrate. CYN inhibited the activity of XOD in a dose dependent manner (IC50 : 26.9 M). The enzyme kinetic studies indicated XOD inhibition in the presence of CYN occurred in a non-competitive manner. The KM and Vmax values of free XOD enzyme were determined as 0.13 mM and 0.0077 U/mg protein, respectively. The apparent Vmax value was 0.0025 U/mg protein in the presence of 100 M CYN, while nearly the same value was determined for KM . KI value of XOD inhibition by CYN was detected as 133 M. In the light of these results, Caulerpa spp. can be considered as important natural resources of bioactive secondary metabolites. doi:10.1016/j.copbio.2011.05.346
