University Dept. of Medicine, Royal Infirmary, 10 Alexandra Parade, Glasgow.G31 2ER, U.K. Studies on heparin fractions have shown a molecular size dependency of the anticoagulant activity. We studied the effect of CY222, a heparin fragment with a mean molecular weight of 2,500 daltons, on the coagulation mechanism and platelet function of normal volunteers after i.v. injection. In addition we examined the reversibility of these effects with protamine sulphate. Standard heparin and saline were used as positive and negative controls. CY222 in doses ranging from 5,000 - 25,000 anti Xa units (500 2,500 U USP) produced a significant increase (p (0.001) in anti-Xa activity by both clotting and amidolytic assays maximal at 20 minutes after injection. There was also a significant (p
