Effect of recombinant human Tumour Necrosis Factor on contractility and prostaglandinsynthesis of pregnant human myometrium U.Karck and H.P.Zahradnik, Universit~tsfrauenklinik Freiburg, Hugst~tter StraBe 55, 7800 Freiburg, FRG We tested the influence of rhTNF on the spontaneous contractility and prostaglandinsynthesis of pregnant human myometrium obtained during primary caesarian section. Small myometrium stripes were prepared and used in a superfusion system. Contractions were recorded and prostaglandin concentration in the superfusate was measured by radioimmunoassay. We could demonstrate that in contrast to speculations in the literature even repetative exposure to rhTNF alone does not lea to increased contractility of myometrium during the observation periode of 12 hrs. Prostaglandinsynthesis remains unaffected during this time, a trend to an increase of 6-k-PGF. , the metabolite of the myometrial relaxant and predominantly formed PGI2, was not statistically significant. These data show that fo the suggested influence of TNF in premature labor either long exposure is required or other prostaglandin synthetising tissues like amnion or decidua might be involved. The immune response of sow's endometrium on diluted boar's semen and the significance of uterus antigens for reproduction stimulated was investigated. For local immune response study to each of 29 sow's the 7.I09 sperms wls introduced. Pot insemination results study to 279sows 2,5-4.I0 v live sperms only or to I4 sows together with 5.I0 at 50°C heating dead sperms was introduced into uterus. After sows artificial insemination was seen activated the local immune response: antisperm antibodies in uterus mucus appeared, the macrophages number in endometrium increasing from 9 to 74 per mm 2 and total lymphocytes from 968 to ~331. The decreasing of live sperm number from 5.I0~ %o 2,5-4.I06 reduced the farrow number %o 35,5%, but after addition dead sperms the farrow number was reestablished to 62,9%. The results suggested that sows artificial insemination stimulated local immune response into endometrium tissues. Th~s event is useful for embryos livability.
