Aspergillus oryzae transformants which produced Bacillus licheniformis thermophilic tr-amylase (optimum temperature, 76°C; optimum pH, 9) were bred using A. niger no. 8AN promoter. When an A. oryzae transformant was grown on rice koji and wheat bran
308-312. 1998. The effects of a packed bed in the downcomer on the average gas holdup and liquid circulation time in a bubble column with a draft tube were experimentally investigated for various gas flow rates and packed bed heights. Gas was dispersed into the inner section of the draft tube or the annular section between the column and the draft tube. In both types of gas dispersion, the presence of the packed bed caused a slight decrease in the average gas holdup and a large increase in the liquid circulation time compared with the values obtained in the absence of a packed bed. As the gas flow rate was increased, the average gas holdup also increased and the liquid circulation time decreased. The degree of reduction in the liquid circulation time was dependent on the type of gas dispersion and the packed bed height. On the basis of these observations, empirical equations predicting the liquid circulation time were obtained.
koji, 15Og total rice was liquefied by koji representing 13% of the total rice and by extraction of log wheat bran koji, respectively. When small-scale brewing tests with liquefied rice and the transformant koji or extraction of wheat bran koji were carried out, the brewing profile and sake analysis values were similar to the results of tests carried out using a control enzyme (CP-40). The results indicate that A. niger no. 8AN promoter can be applied to the production of heterologous proteins such as B. licheniformis thermophilic tr-amylase in
