HUMAN KJ29 CELL LINE, ! S O L A ~ FROM A KIDNEY ADENOCARCINON~: MORPHOLOGICAL ASPECTS A~]D EFFECTS OF CHEMICAL INDUCERS. Cattedra di Ematologia - Universit~ degli Studi di Ferrara, Ferrara, Italy A cell line named K J29 has been isolated from adenoesa~cinoma cells of proximal convoluted tubules. These cells proliferate in Y~M, i(~/0FCS, maintaining an epithelial aspect. Approximately after i0 days of culture, vesicles and pseudotubules appear from the adherent cells, as shown in Fig. i.

the KJ29 cells could be modified "in vitro" we have analyzed how these cells respond to different chemical inducers, such as 12-0-te tradecanoyl-phorbol- iS-acet ate (i00 ng/ml), protamine (200 ~g/ml), dimethylsulphoxide (DMSO , 2%), butyric acid (BA, 1.2 rnM) and examethylene-bisacetamide

(HMBA, 5 mM). DMSO , HM~A and BA inhibit both K J29 cell proliferation and organization, producing a more flattened configuration of the cells (see Fig. S A). In addition, the 12% po!~acrylamide-SDS • ~w . . gel electrephoresls of S - m e t h m o m n e labelled proteins shows a different pattern in BA treated with respect to untreated cells (Fig. 3 B).

In conclusion this new human kidney cell line, retaining both tumoral features and differentiated properties of kidney cells, may prove a useful model to study the mole c u l a r events responsible of kidney cell proliferation and differentiation.

Other kidney features are present in these cells such as a) the expression of two antigens typical of proximal tubules (L. del Senno, manuscript in preparation); b) the formation of "domes" (2) and of vesicles which increase by inside water accumulation.
