~ysyl oxidase initiates the cross-linking of collagen and elastin by catalyzing the formation of the lysine-derived aldehyde. we Investigated the localization of lysyl oxidase in normal human using skin and indirect immunofluorescence alklaline phosphatase anti-alkaline phosphatase method with monoclonal anti-human lysyl oxidase antibodies. Fine filamentous or granular positive lmmunoreactions were noted on or among collagen or elastic fibers. Basal cells, endothelial cells and sweat glands also tested positive as well ds flbroblasts and arrectores pilorum.

582 PRE-OPERATIVEDIAGNOSISOF MALIGNANTMELANOMA BY THE NEEDLE ASPIRATION-FLUORESCENCE METHOD RAZUOCHINO,HIDES FUJITA, AKIHIKO SHIBATA AND TAKAFUMI MORISHIMA Departmentof Dermatology,NihonUniv.School of Medicine,Takyo A diagnosis of melanoma from clinical features as d definitive diagnosis before surgery is difficult. The purpose Of this study is whether it is Dassible to diagnose melanoma before operation by the aspi;ation-fluorescence method. Aspirated specimens were Caken from 25 primary melanomas, 11 cutaneous metastarir lesions, 2 lymph nodes of metastasis and 290 non-melanoma tumors. Fluorescent melanoma cells were easily detected in all melanoma cases. and all cases were diagnosed as melanomas before operation.In contrast,no fluorescenttumor cells were observed in non-melanoma cases except in 30 pigmented tumors. But, the fluorescence was weak and shape of the cells were dendritic, consequently the tumors were diagnosed as non melanoma. This study has confirmed that this method is useful for pre-operative diagnosis of melanomas.

When the Bm cDNA [JBC. 1988) was transferredinto BlG-FLO. invasiveness tn vitro dOOreased aOa min stress fibers increased (FEBS.1990). In this study we transferredthe 81 CDNA into highly metastatic B16-BL6 cell line in addition to B16-FlO in order to investigateeffects of Brn actin on metastasis. r&tastasis axoerimentsin V~VOana WOsterO blot analysis showed that aetastasiswas reduced in transfectantsexpressing Bn. These results suggest that Br actin Plays an Importantrole to reduce metastatic ability in q~oUsa 016-melanoma.
