INHIBITION OF EDRF/NO PRODUCTION IN RABBIT AORTA BY SERUM PURI FIED FROM FACTOR/S IN HYPERTENSIVE PREECLAMPTIC PARTURIENTS A.Pinto,R.Sorrentino,L.Sautebin,P.Martinelli*,T.Guerritore*and L-Sorrentino Pharmacology, Faculty of Pharmacy and *Inst. Experimental of Gynaecology, Obstetrics and Physiology of Human Reproduction -11 Faculty of Medicine and Surgery, University of Naples "Federico II" In previous work we showed a marked reduction of EDRF/NO from umbilical endothelial cells of pregnancy-induced hypertensive parturients. This results let us to hypotize a generalized reduction of EDRF/NO production in the mother circulation, possi ble due to the presence of an endogenous inhibitory factor/s.To test this hypthesis we have fractionated, by gel filtration (Sephadex G 200), the serum of preeclamptic parturients and found two fractions named F, and F6 that significantly inhibited (56.6+8.5% and 72.656.3s respectively, p 0.01) NO generation mediated by ACh (0.01-l uM) in rabbit aorta rings-The apparent m-w. of F was greater than 400 kd and the m-w. of F was minor than 1 b kd. Further studies are in progress to be Eter characterize these inhibitory factors.

Spm-SP inhibited collageninduced relaxation of aorta rings, mediated platelet aggregation and induced only a slight inflammatory response in the rat hind paw failed to contract the isolated rat stomach strip and duodenum. Since the antagonist of nitric oxide biosynthesis, L-NAME effectively we conclude that Spm-SP retains counteracted Spm-SP effects, to receptor(s) only the specific property of SP to bind involved in the transduction for the signal of nitric oxide release.
