absorbance at 615 nm (E = 5700 M-1 cm-l), Type 2 copper depleted lactase was prepared according to the method of Graziani et al (l-3). Its copper content was determined spectrophotometrically with biquinoline. The two, independently prepared samples of T2D lactase studied in this work contained 2.8 and 2.9 Cu ions per mole protein, respectively. In view of changes caused in the molar extinction at 280 nm of T2D lactase, protein concentration was determined by the Lowry method. This showed that while essentially no change occurs in the intensit of the 615 nm band, that at 280 nm increases to Q99700 M- 4: cm-l (6). T2D lactase preparations were oxidized by excess ferricyanide which was removed by extensive dialysis or gel filtration. Protein concentrations examined ranged from 23 to 70 PM. CD spectra were measured in 1 cm cuvettes on a Roussel Jouan Dichrograph III. Data were collected by means of a Tektronix minicomputer averaging 200 measurements per nm.
