Abstracts * A CYTOGENETIC STUDY ON LEUKEMIA USING AJ5 HIGH RESOLUTION TECHNIQUE Yang Aide.Fei Hongbao,Fu Tao,Gong Weilong, Institute o~ Hematology/Cytogenetics,Xiehe Hoepital,Tongji Medlcal University,wuhan0 China 55 cases of diffrent kinds of leukemia were studied with FDU-syncnronization technique anu ~8(87.5~) O f t h e m w e r e 1 o u n d to h a v e a b normal karyotypes. The incidence of chromosomal aberration was much higher in prometaphase than that in mezaphase. Some minute changes which coul~ nee be identified ar the s t a g e 0£ 2 2 0 b a n d s w e r e s h o w n b y u s i n g high resolution technique. One case of CML was considered re have an atypical Ph,t(9~22), ar metaphase and was proved ¢o be a complex cranslocation of t(9;22~I0) at the stage of more than 500 bands. Another case of CML also foun d tO h a v e a c o m p l e x translocation t(9~22;17). Two infants were diagnosed as Ph negative CML a t f i r s t b u t Z u r n e d o u t to b e a m y e l o d y s p l a stic syndrome after the demonstration Of t(gp llq) and re-examination of bone marrow cells. Abnormal karyotypes were founa in 2 of the 3 patients with histocytic leukemia. One had trisomy of lq and the other had multiple chromosomal changes, t(8~21),t(15;17) and Ph were noted in M2,M3 and CML respectively and the bre~kpoints of chromosomes were determined more precisely by using this method.

The low m i t o t i c index of f r e s h s o l i d tumor specimens impedes t h e a c q u i s i t i o n of c y t o g e n e t i c s a t d i a g n o s i s of m a l i g n a n c y . Premature chromosome c o n d e n s a t i o n (PCC) i n i n t e r p h a s e c e l l s o f f e r s a s o l u t i o n once an e a s y method and a v a i l a b l e components a r e a s s u r e d . We r e p o r t h e r e t h e a v a i l a b i l i t y of a Chinese h a m s t e r o v a r y (CHO) h y b r i d c e l l l i n e t h a t has a f u s i o n and PCC i n d u c e r p o t e n t i a l far g r e a t e r than p r e v i o u s l y described. Cell l i n e R103, a CRO h y b r i d c o n t a i n i n g some chimpanzee chromosomes has a n e a r o c t a p l o i d number of 70 t o 90 chromosomes. R103 c e l l s grown o v e r n i g h t and t r e a t e d 3 h o u r s w i t h c o l c i m i d a r e f u s e d witb c o l l a g e n a s e d i s a g g r e g a t e d s o l i d tumor s p e c i m e n s . F u s i o n by Sendai v i r u s and e x p r e s s i o n of PCC's r e q u i r e s two h o u r s p r i o r t o t r e a t m e n t as a r o u t i n e c y t o g e n e t i c p r e p a r a t i o n . HeLm c e l l s have been used by B r u d e r l e i n e t a l . (Hun Genet 73:44-52, 1986) and Hittelnan et al. {Cancer Genet Cytogenet, 30:301-312,1988) as an inducer cell. The former authors used a 48-hour preparation with electric field pulse on in situ preparations while the latter used 48-hour preparations and a pressure bomb for collecting mitoses. The mitotic shake-off cells of R103 have been fused with diploid lymphohlastoid cell lines,
h y p e r d i p l o i d leukemia c e l l s , HeLa, h y p e r d i p l o i d SCLC c e l l l i n e s and m u l t i p l e c o l l a g e n a s e t r e a t e d tumor c e l l s from f r e s h s p e c i m e n s . Condensed c h r o m a t i d s a r e e a s i l y d i s t i n g u i s h e d from CHO h y b r i d m i t o t i c chromosomes. For use i n t h e s e f u s i o n s , m i t o t i c CHO c e l l s can be p r e s e r v e d a t -70C i n DNSO, making t h i s p r o c e d u r e r e a d i l y a v a i l able.
