215. RELATIONSHIP OF THC INDUCED IMMUNOMODULATION AND IL-2 ACTIVITY IN MURINE SPLENOCYTES AND LYMPH NODE CELLS. Y. Nakano, S. Pross, S. McHugh, C. Newton, and H. Friedman, Univ. So. Florida Col. of Medicine, Tampa, FL 33612. The effects of THC (delta-9-tetrahydrocannabinol) on young and adult murine spleen and lymph node cells were investigated. This study shows an organ specific difference in the immunomodulating effect of THC. Thus, THC inhibited the CD3 induced proliferation in adult lymph node cells, but upregulated the response in the adult spleen. Interestingly, THC did suppress the CD3 induced proliferation in young spleen cells. This difference in the response of adult and young spleen cells reflected a difference in response of Ly2 + cells to the CD3 and THC. IL-2 activity was analyzed from these experiments. It was found that CD3 alone increased IL-2 activity in both young and adult lymph node and spleen cells. THC upregulated IL-2 activity in adult spleens in contrast to the young spleens. This possibly reflects the contrasting effects of THC on Ly2 cells in these two age groups. Supported by NIDA DA-06385.

216 EFFECTS OP BCG VACCINE ON FUNCTIONAL PROPERTIES OF ~ C R O P H A G E S V.I.Seledtsov, G.g.Seledtsova Institute of Clinical Immunology, Novosibirsk, USSR The effects of Bacillus Calmette Guerin (BCG) vaccine on functional properties of tioglicolate-stimulated peritoneal macrophages (M@) of (CBA~C57BL/6)F~ mice was studied in vitro. BCG in 10-50 ~ g / m l concentrati6ns was shown to inh~bite Fc-dependent phagocytosis. Supernat%nt (10-30%), containing ~ - a c t i v a t i n g f a c ~ (~AF), from mice mixed lymphocyte culture, exerted an analogous inhibiting effect on Pc-phagocytosis. However, in the presence of high NAF concentrations (more 10%) BCG (25-50 /,g/ml) stimulated Fc-phagocytosis. BCG could be a second additional signal for activation of antitumor function of Mq. BCG (0.1-10/,g/ml) 2-5 times increased NAY-induced cytotoxic effect of M~ on P815, L1210, K2 cells. BCG itself even in 10 ~g/ml concentration did not induce antitumor M~ activity. Acting on M~, BCG did not increase the adhesion of tumor cells to M9 while NAF had that effect. Folyclonal antibodies (mainly IgG) against BCG wer& able to abrogate BCG-dependent antitumor activity of Mq. Thus, ~AF and antibodies against BCG can determine the final result of BCG-M~ interaction.

Gumilevsky Institute of Pharmacology, Tomsk, USSR The study was to estimate the production of IL-I, IL-3, CSF by mouse bone marrow cells during regenerating processes after cyclophosphamide injection. The top of decreasing of myelocariocytes was noted at day 3. At next days the count of bone marrow cells increased especially by means of immature granulocytes. However, in this time the number of CFU was deep decreased and was low level of CSF from adherent bone marrow cells. The synthesis of IL-I by adherent cells increased at I day after injection of CY with the top on 3 day. At another days the level of IL-I production decressed, but up to the day 10 was still higher then in control. The IL-3 activity appeared at supernatsnts of nonadherent cells at I-2 day after CY treatment. The maximum production was registrated st 6-7 day. It was proposed the availability of shunt mechanisms of primitive hemopoietic cells differentiation, activated after action of myelotoxic agents and marrow mononucleares synthesis may be early sygnal for this processes of regenerating bone marrow after cytostatic therapy.
