PROGESTERONE CONCENTRATION IN ALLOGENEIC SKIN TRANSPLANTED ORTHOTOPICALLY AND INTO THE CHORIODECIDUAL JUNCTIONS OF RATS AND ITS EFFECT ON SKIN A L L O G R A F T SURVIVAL. Jimmy O. Sio, James F. Progesterone (P) has been shown to interfere with the elicitation and the expression of immunity in vivo and in vitro. To address progesterone's involvement in the immunoprotection of the fetal-placental unit serum and tissue P concentrations in skin allografts, determined by radioimmunoassay, were compared with skin allograft survival in Fischer (FI) rats receiving: A. (FI x DA) F 1 hybrid orthotopic everted tail skin allografts placed over silastic tubes containing 50 or 100 mg of P and grafted subcutaneously; B. (FI x DA)F 1 hybrid skin allografts placed subjacent to each fetal-placental unit of FI females gestating FI conceptuses so that the dermal surfaces healed in on the decidua and the squamous epithelium was juxtaposed to the trophoblast. It was found that: 1. Tissue concentrations of P in orthotopic extrauterine skin grafts of approximately 100 ng/mg were necessary to achieve marked prolongation of allograft survival. 2. This correlated with serum levels of 15 to 20 ng/ml of P. 3. Skin allografts demonstrated impeccable viability with no signs of rejection in the choriodecidual junctions. 4. P concentrations in these choriodecidual allografts was equal to or less than 1 ng/mg of tissue. 5. The serum P concentrations on day 21 of pregnancy, the day of sacrifice, were more than twice those achieved in the above experiment (45 to 60 ng/ml). As a control P concentrations in tissue removed from the uterine implantation site, sites not involved in implantation, placentas, and tissue removed from the pseudopregnant uterine horn on day 21 of pregnancy revealed values consistent with those found in the choriodecidual skin allograft (0.8 to 1.2 ng/mg of tissue). These experiments highlight that the prolongation of allograft survival seen in the choriodecidual junction of the gravid uterus of the rat is not due to the action of P alone, for this concentration is consistently 100 fold less than that required to achieve significant prolongation of allograft survival in non-uterine sites.
