Secretion of soluble cytokine receptors has been s u g g e s t e d as a m e c h a n i s m for t h e r e g u l a t i o n of c y t o k i n e a c t i v i t y in vivo. I n c r e a s e d s h e d d i n g of s o l u b l e T N F - R is a s s o c i a t e d w i t h n o r m a l p r e g n a n c y and i m m u n o h i s t o l o g y has d e m o n s t r a t e d the p r e s e n c e of T N F - R in b o t h t h e p l a c e n t a a n d d e c i d u a . T h i s s t u d y w a s d e s i g n e d to see w h e t h e r TNFR w e r e s h e d f r o m p l a c e n t a l or d e c i d u a l tissue. Immunoassays for soluble TNF-R were performed as previously described [I] . M e d i a conditioned by incubation with decidua or chorionic villous tissue was f o u n d t o c o n t a i n s i g n i f i c a n t a m o u n t s of s o l u b l e T N F - R . In t h e f i r s t t r i m e s t e r , decidual tissue produced more p55 TNF-R than the placenta. H o w e v e r at t e r m , h i g h e r l e v e l s of p55 T N F - R w e r e found in c o n d i t i o n e d m e d i a f r o m c h o r i o n i c villi. O u r r e s u l t s s u g g e s t t h a t p l a c e n t a l s h e d d i n g of T N F - R into the maternal circulation may play a role in r e g u l a t i n g T N F - a a c t i v i t y in p r e g n a n c y . i.

Department of Obstetrics and Gynecology, Saitama Medical School, Moroyama, Saitama, Japan. Proliferating cell nuclear antlgeu(PCNA) Is one of the markers for cell proliferation. The immunohlstochemical localization of PCNA Is Investigated in placental tissues at the first, second and third trimester of pregnancy with no complication and placectae with toxemia of pregnancy. PCNAwas positive on nuclei of cytotrophoblasts(CTbs) of free and anchoring v i l l i In early stages of gestation. Nuclei of syncytiotrophoblasts were not stained. PCNA positive cells gradually decreased as progress of gestatlonal age. In cases with toxemia of pregnancy, PCNApositive CTbs existed more than normal placentae at the same stages of pregnancy. Although PCNApositive cells decreased In the central areas of lntervlllous thrombosis or the parts with infarction. Nuclei of flbroblasts and capillary endothelial cells sometimes seemed to be PCNA positive. The increase of PCNApositive cells in toxemlc placentae seemed to be one of the reparlng mechanisms of placental tissue.
