We describe here a mo&kation of the Technicon technique E8j F&rthe MC II P AutuAnalyzer. KoeM and MandeEE9f had akeady modified thk technique by programmingan automatic temperaturechange for tie columns and by rumzing two cohmns in paraUel.thus daubLingthe capacity of f&e in&ument and permittingsimuhaneous analyses of two different sampIes_Our modification substitartesthree buffers for the seven previously used, and akws the separation of 40 amino acids and ninhydiin-positive substances of in&rest in human biology from as little as 50 crfof plasma or urine.

TwocoIusms(478 X 5 mm)n;ount.&inparaEeE and fed byt;FVohigfr-presszlze pumps, which are tomperahre.contro&d by turo EG&e water-bafhs, are each packed with a 4fcm bed of ChromobeacfsC3 resin bezds (p&&e diameter IQ -1. These coEumnsare eked to the XC II P Teclmicon system, consisting of a P 111 pump, a perishkic valve and a programmer,a 95” waterbath with a double coil, two slngk-cbannel calorimetersfor tie AutuAndyzer II. equip_& with a set of interference Nters at 4BO nm, and a double-pen recorder.

Reagents

The ninhydrin reagents are prepared as recommended by Technicon, ninhydrin and hydrazine suIphat.2being mixed as required. The pH of the ninhydrin reagent is adjuskd to 6.30 for increasedsensitivit;y[IQE. Afeer they have been used, the resins are regenerated in 0.3 159Lithium hydroxide sokkion end re-equilibrakd ~6th the first buffer at pH 2.15. The Iithium citrate elution buffers E3, El, 221 are prepared as shown in T&k I. PlaSma or urke saznpfesare deproteinized with 5% or 2% suEphosalicylic acid, respectively. They are then dikt4& ~6th an equal vohme of the sampledihtior: @K 2.20) des&M by i3ensanet al. &3].The final pH of the sampleis 2.20.
