During physical exercise hemostatic system is significantly modified (increase activity and levels of FVIII, fibrinolysis activation), but a clarified definition of these modifications with respect to course of physical activity doesn’t exist. Aim of this study was to evaluate a possible clotting activation during a physical standardized exercise by measuring the following parameters: prothrombin activation fragment (F1+2, ELISA, Behring Institute), thrombin-antithrombin complex (TAT, ELISA, Behring Institute), fibrinopeptide A (FPA, ELISA, BMI) and D-dimer (D-D, ELISA, BMI). Moreover, plasma levels of antithrombin III (AT III, amidolytic method, Behring Institute) were determined, to evaluate a possible consumption. Ten healthy male volunteers (mean age: 28*3 yrs), not usually engaged in any sport, who underwent exercise test using the modified Bruce protocol were examined. Ventilatory volumes and gas exchanges were measured by computerized system, that allows breath by breath analysis. To determine the stage in which each subject reached the anaerobic threshold, three days before the study, a first exercise test was performed. Then, in the experimental day blood withdrawals were executed to measure the different parameters: 1) before exercise; 2) at the end of the stage preceding that in which anaerobic threshold was reached; 3) at the maximal exercise; 4) 30 min after stopping exercise. The activation of fibrynolitic system was evaluated in 15 healthy males volunteers (age 28k4 years) who, in consecutive days, underwent two treadmill stress tests according to the modified Bruce protocol or to the Norton protocol so to perform the same work load in aerobiosis or in anaerobiosis. The anaerobic threshold (AT) and the peak oxygen consumption (V02 max ) were determined in each subject with a Sensor Medics Unit 400 two days before the study was performed. Samples for determination of fibrynolitic parameters were obtained in supine position before exercise, at maximal exercise and 30 min after stopping exercise; during the Bruce protocol a fourth sample was got at the end of the stage preceding the one in which the AT was reached. Plasminogen activity, euglobulin lysis time (ELT), t-PA ag and act, PAI- ag and act and DDimer were evaluated; adrenaline was assessed by HPLC with electrochemical detection. During the exercise in aerobiosis a slight decrease in ELT and a small increase in t-PA were observed.
