METHODS: Expression of apoptosis-related genes was examined by PCR-Array assay. RESULTS: Expression of BAX,P53 and P73 increased significantly whereas the expression of TRAF and BFAR decreased in SiRNA-Ku70 A549DDP cells compared to A549DDP cells (P< 0.05) . CONCLUSION: These results show that Ku70 might play an important role in drugresistance in A549DDP cell line. Silencing of Ku70 can be attributed to the elevation of apoptotic genes- BAX, P53 and P73 expression; reduction of anti-apoptotic genes TRAF and BFAR. This study suggests that silencing of Ku70 might reverse the drugresistance in A549DDP cell line. CLINICAL IMPLICATIONS: Ku70 might be a new target for the drug-resistant reversion.
