Adult rat cardiomyocytes (ARC) in culture undergo extensive changes in morphology and in expression of contractile proteins similar to that found in fetal heart cells. In contrast to fetal cells, though, ARC synthesize and accumulate mitochondrial creatine kinase (Mi-CK) and show, by using antibodies against this protein, two types of mitochondria: large, cylindrically shaped randomly distributed mitochondria exhibiting a strong signal for Mi-CK, and small round mitochondria organized in rows parallel to myofibrils and giving a much weaker signal. EM investigations revealed in the large mitochondria paracrystalline inclusions similar to those described in vivo in mitochondrial myopathiesln ARC these inclusions were specifically decorated by gold-coupled Mi-CK antibodies. Both, large mitochondria and their inclusions disappear upon addition of 20 mM cmatine to the cmatine-free culture medium. We suggest ttutt a low intracellular total creatine level (rtratinc plus phosphncreatinet metabolic stress and the high concentration of Mi-CK within the inclusions may compensatory effect. Furthermore ARC in culture could serve as an in virro model to study

Compatibility and oxygenation ability assessment of 70 g/l stmma-free human hemoglobin sokttions pyridoxylated (PLP) or not, and purified or not, was carried out using microvascular perfusion of intestine of rats. Mechanical and ultmstructural changes in the organ and the arteriovenous difference of PO2 were compared to those obtained with current plasma substitutes : albmnin. gelatine and dextran. Overlapping epidtelium keeps its structure in the presence of hemoglobin, whereas it is partially or totally destroyed with the plasma substitutes. The intestine. irrigated by the hemoglobin solutions shows efficient peristalsis, but this totally disappears with plasma substitutes. For similar arte-tial PO2. for all the solutions, the arteriovenous difference was of 110 mmHg on average for the hemoglobin solutions, whereas it never exceeded 60 mmHg with the plasma substitutes.
