Methods: A novel 3D co-culture model was constructed by HUASMCs (human umbilical artery smooth muscle cells) and HUVECs (human umbilical vein endothelial cells) in Transwell chambers. Secretions and transcription of VEGF, t-PA, NO and IL-8 from HUVECs and VCAM-1, MMP-2 and MCP-1 from HUASMCs were detected by Elisa and Real-time PCR among four groups (leptin, leptin + adiponectin, leptin + losartan and control). Expression of AdipoR1 (adiponectin receptor) or AdipoR2 of HUVECs was inhibited using RNAi technology. Changes of intracellular Ca2+ in HUVEC were detected by Fluorescent probe Fluo-3Am. Changes of p-AMPK/AMPK, p-STAT3/STAT3 were tested by western blot. Results: VEGF and NO were increased by leptin and not affected by adding adiponectin or losartan. IL8 were unaffected by leptin but increased by adding adiponectin (P b 0.01). VCAM-1 was increased by leptin but reduced by adding adiponectin (P b 0.01). Results of real-time PCR were the same as ELISA. Receptors of HUVECs could be effectively inhibit by AdipoR1-siRNA and AdipoR2-siRNA with inhibition rates of 71.83 ± 1.45% and 74.89 ± 1.12% (P b 0.01). The effect of adiponectin in increasing IL-8 and reducing VCAM-1 was diminished by the depression of AdipoR1. Adiponectin increased the concentration of intracellular Ca2+ of HUVECs. p-STAT3 was activated by leptin while p-AMPK was activated by adiponectin. Increasing of IL-8 and reducing of VCAM-1 by adiponectin could be inhibited by compound C and partially inhibited by PD98059. Conclusion: Cell growth and secretion in 3D co-culture model were different from the plane culture. Crosstalk between HUVECs and HUASMCs was mediated by IL-8 and it reduced secretion and transcription of VCAM-1via ERK pathway. By activating AdipoR1, adiponectin increased the concentration of intracellular Ca2+, and exerted partial anti-inflammatory effects against Leptin though the AMPK pathway. Adiponectin might also increase the secretion of IL-8 in HUVECs to reduce VCAM-1 from HUASMCs.

Federation (IDF 2005), 133 consecutive Chinese hypertensive subjects were divided into two groups: essential hypertensives (EH group, n= 36) and those with metabolic syndrome (EH + MS group, n= 97). Biochemistry assay, plasma vWF, 24 h urine albumin excretion (UAE) and morning urine albumin/creatinine ratio (ACR) were measured. The association of vWF and MAU with MS was analyzed. Results: Waist circumference, body mass index, triglyceride (TG), low density lipoprotein cholesterol (LDL-C), plasma vWF and UAE were increased (P b 0.05 or P b 0.01, respectively), while high density lipoprotein cholesterol (HDL-C) was decreased (P b 0.01) in EH+ MS group, as compared with those in EH group. UAE was positively correlated with ACR (r= 0.707, P b 0.01). vWF was not correlated with UAE or ACR (r= 0.079 and 0.052, PN0.05, respectively). Logistic regression showed HDL-C, TG, LDL-C,vWF and UAE were associated with the development of MS in hypertensive patients (standardized coefficients: −0.825, 0.63, 0.339, 0.331, 0.371, P b 0.05 or P b 0.01, respectively). Two ZFP36 polymorphisms were associated with body weight, body mass index (BMI), as well as glucose and LDL-cholesterol levels in a cohort study. ZFP36 gene expression in omental adipose tissue, but not in abdominal s.c. fat, may offer partial protection against the development of insulin resistance and diabetes. Therefore, we investigated in an association study of Uygur subjects at high risk for obesity whether genetic variation within the ZFP36 gene locus contributes to MetS phenotypes. Methods: We genotyped 1021 subjects (647 women, 374 men) for two single nucleotide polymorphisms (SNPs) tagging 100% of common variants (MAF N 0.05) within the ZFP36 gene locus with an r2 ≥ 0.8. All subjects underwent questionnaire, physical examination, measurement of lipid levels, TNF-α in the plasma. Results: After appropriate adjustment and Bonferroni correction for multiple comparisons, none of the rs251864, rs3746083 SNPs was reliably associated with MetS. Multiple logistic regression indicated age [OR (95%CI): 1.02(1.00–1.03), P = 0.02], being female [OR(95%CI): 2.16 (1.61–2.89), P b 0.001] and alcohol intake [OR(95%CI): 2.97 (1.18–7.47), P = 0.02] were significant factors.
