The average concentration of glucocorticoid receptor in various intracranial lesions was as follows: cerebral metastases > meningiomas > acoustic neuromas > gliomas > peritumoral brain tissue > contused brain tissue. The clinical response to dexamethasone in brain edema associated with various intracranial lesions is to some extent parallel to the glucocorticoid receptor concentration. The study on the relationship between computed tomography (CT) findings and in various intracranial tumors showed glucocorticoid receptor concentrations that some correlation existed between the glucocorticoid receptor level and CT contrast enhancement. This indicated that glucocorticoid receptor interaction is of importance in the regulation of the vascular permeability of tumors. Recently, interest has centered on sex steroid receptors, i.e. ER, PR, and AR, in intracranial tumors because more specific treatments of patients with malignant glioma and recurrent and/or unoperable meningioma are needed, and because epidemiological and clinical data have indicated some sex hormone sensitivity. We therefore assayed adult tumor biopsy specimens systematically for cytosolic receptors, i.e. ERc,PRc and ARC, using respectively (3H) R2858, R5020 (or RU27987) and R1881 as radioligands, and in some cases for nuclear receptors (Rn). R5020 binding sites were also measured in meningioma cell cultures. In glioma (n = 20), ARC (mean Kd: 2nM) and ARn were detected, but at low levels, in 90 and 60% of samples respectively; ERc was detected in a few cases only and PRc was apparently absent. In meningioma (n = 46), ERc, PRc and ARC were found in 33, 93 and 87% of samples respectively (Kd of the nM order). ER levels were low in the cytosol and/or the nucleus whereas ARC and PRc (and PRn) levels were moderate to high (PR levels up to 25000 fmol/gT) enabling biochemical characterization of AR and PR. R5020 binding activity was detectable in meningioma cell cultures. The relevance of AR in glioma and of AR and PR in meningioma is discussed in relation to tumor cell proliferation and differentiation.
