52042~ PROCESS FOR THE REDUCTION OF STAR ACTIVITIES Frank Laue, Waltraud Ankenbauer, Gudrun Sehmitz, P assigned to Boehringer Mannheim GmbH The present invention provides a process for the reduction of non-specific star activities in the case of the specific cleavage of desoxyribonucleic acids by incubation with a restriction endonuclease in an appropriate buffer, wherein to the incubation batch is added an antibiotic which binds to the D N A on or near the star sequences of the enzyme but not within the specific recognition sequence of the restriction endonuclease.

The recombinant poxvirus is modified by disassociating the internal core from outer membranes of the poxvirus. The D N A codes for and expresses a gene product in a cell cultured in vitro and in a host in vivo without productive replication of the virus in the cell or in the host. What is also described is a vaccine containing the modified recombinant poxvirus for inducing an immunological response in a host inoculated with the vaccine.

The procedure does not require deproteination and yields clean DNA that is suitable for both PCR and Southern blotting. It can be utilized on a small scale without homogenizing the tissue. These features also facilitate automated screening of plant tissue samples, one of the labor-intensive techniques in molecular biology. This method is also adaptable for use in the field.
