Book Review Techniques in Immunocytochemistry, Vol. 2, edited by G.R. Bullock and P. Petrusz. Academic Press, London, 1983 (300 pp.) £25.-/$41.50 This book complements Vol. 1 of the series, published in 1982 and reviewed in this journal in 1983 (Vol. 58/3). It is focused on certain recently developed methods and their applications. The first and the final chapters deal with different aspects of using colloidal gold-labelled antibodies for the detection of cell-surface and intracellular components. Scanning electron microscopy combined with the immuno-peroxidase and other labelling methods is the subject of 2 contributions. The use of the biotin-avidin complex in coupling antibodies to enzymatic labels is described briefly in one paper and discussed at length elsewhere. Another chapter is devoted to the review of techniques used in the production of monoclonal antibodies and their application as immunocytochemical reagents. A topic of methodological interest is also the use of lectins, labelled directly or through an antibody, to identify carbohydrates in paraffin sections of tissues. Among the applications of immunocytochemical techniques reviewed herein is the localization of estradiol receptors in estrogen target tissues. A comparison is made between the results of autoradiography using the labelled hormone and those of staining tissue sections with enzyme-labelled anti-hormone or anti-receptor antibodies. Further one should mention the descriptions of immunofluorescence studies on double-labelled neurons at both the light and electron-microscopic levels, and the localization of ACTH and other anterior pituitary hormones in gland sections and in dissociated cells employing the avidin-biotin enzyme system. The volume, like its predecessor, is profusely illustrated with photomicrographs and.schematic diagrams, and provided with a comprehensive subject index.
