4829004 ROLLER
BOTTLE
SYSTEM
James Varani, David Solomon, William J Hillegas, David L Melmoth assigned to The University of Michigan; Solohill Engineering I An insert for a roller bottle of the type used in cell culture for vaccine manufacture is formed of a polymeric resilient material whereby it can be bent or rolled so that it can pass through the opening of the roller bottle. Once the insert is inside the bottle, it is released so that its resilient characteristic causes it to unbend or unroll. The insert is wider than the diameter of the roller bottle, and therefore it is urged against the interior wall of the roller bottle. Apertures and/or notches are provided in the insert to increase the turbulence of a fluid and to decrease the fluidic shear forces in the roller bottle as it is rotated in conventional operation. A significant increase in productivity is achieved because particles, which may be microcarrier beads, are maintained in suspension, and prevented from sinking to the bottom of the fluid medium where cells cannot grow. The roller bottle system is thereby transformed into a microcarrier suspension culture system.
4829009 NOISE CONTROLLED IMMUNOASSAYS Howard Graves assigned to The Regents of the University of California A method for reducing false positives and to assay background noise in solid phase immunoassays utilizes a matrix coated on a solid support, which matrix contains an effective amount of a noise reduction component and an effective amount of noise balancing component. Optimization of the matrix composition is obtained by measuring parameters for its effectiveness which include sensitivity ratio, noise balance ratio, and signal to noise ratio.
4829012 METHOD FOR IMMUNOLOGICAL ASSAY OF ANTIBODIES OF VARIOUS TYPES IN A LIQUID SAMPLE Cesar L Cambiaso, Pierre Masson, Kraainem,
Belgium assigned to L'Association Internationale gra/a/ but Scientifique dite: Institut International de Pathologie Cellulaire et Moleculaire A method for immunological assay of IgM antibodies against at least one said antigen in a liquid sample containing at least IgM and IgG antibodies, such as a biological fluid, comprising reaction of said liquid sample and of antibodies against IgG, and addition to the so obtained reaction product of the said antigen bound to finely divided particles and of a free said antigen, the amount of lgM being inversely proportional to the amount of unagglutinated finely divided particles.
4829098 IMMOBILIZED BIOMOLECULES AND METHOD OF MAKING SAME Allan S Hoffman, Liang C Dong assigned to Washington Research Foundation A method of biomolecule immobilization is described in which the biomolecule itself or, alternatively, a monomer-conjugated biomolecule, is grafted with free monomer onto a hydrophilic, solid-phase, polymeric substrate which has been pre-irradiated with ionizing radiation. The pre-irradiation step is carried out, preferably at -78 degrees C. in air, while the grafting step is carried out at 0 degrees C. in a substantially oxygen-free atmosphere. The technique is applicable to immobilization of a wide variety of biomolecules, such as enzymes, catalysts, hormones, lectins, drugs, vitamins, antibodies, antigens, nucleic acids, D N A and R N A segments, pesticides, dyes and fertilizers. The products may be used for therapeutic or diagnostic applications or bioseparations.
