443 EXPRESSION OF PROLIFERATING NUCLEAR ANTIGEN (PCNA) IN HUMAN PAPULOSIS IS INDUCED BY BOWENOID T. ISHIJIB>, PAPILLOMAVIRUS-16 E7 ONCOPROTEIN. Honda? M. Niimura??, H. Matsushimd, K. Kikuchl and S. Yasumoto?. _A 1Dept. of Dermatology, Atsugl Hospital, Kanagawa. Japan. *Dept. of Dermatology, The Jikei University School of Medicine, Tokyo, Japan. %magawa Cancer Center Research Institute. Kanagawa, Japan. Prewous studies have demonstrated that bowenoid papulosis IS associated with infection of type 16-r&&d HPVs. By using in situ hybridizabon technique wth HPV-16/16 mixture probe, HPV DNA was detected from bowenoid papulosis specimens. In order to examine proliferatwe activities of these cells, expression of PCNA was studied in the same specimens using immunohistochemical method with the mcmoclonal antibody. We detected significant amounts of PCNA accumulation 1” the HP&infected cells. On the other hand, it IS known that HPV-16 E7 gene mduces cellular proliferation of human foreskin kerabnocytes. For checkmg the ability of E7 gene product to Induce PCNA expression, E7 express,o” vector was co-transfected E7 enhanced CAT with PCNA promoter-CAT reporter plasmtd. expression from the PCNA promoter in some cell knes. These data suggested that expression of PCNA I” HPV-16 infected cells could be induced by E7 protein.
