PLACENTAL FUNCTIONSANDCYTOKINES S.Saito, Department of Obstetrics and Gynecology, Nara Medical University,840 Shijo-cho Kashihara-shi,Nara 634,Japan Many cytokine receptors are expressed on the trophoblast cells. We clarified that G-CSF R, M-CSF R (c-fms), SCF R (c-kit) and HGFR (c-met) were expressed on the trophoblast cells, and trophoblast cells produced G-CSF, M-CSF and SCF. These cytokines are produced by not only trophoblast but also decidual cells, mononuclear cells and endometrial gland cells. In this namer. we focus on the olacental function and cvtokines we demonstrated-that in normal pregnancy. ‘As a pathological’events, placental apoptosis through the Fas, TNF/NGFreceptor family, occured in the case of premature separation of the placenta.

[INTRODUCTION] We had reported that high levels of HGF (10%50ng HGF/mg total protein) were detected in placental tissues from early and late stages of pregnancy. In this study we examined the localization of HCF in placental tissues by immunohistolo ical staining and in situ hybridization. [MATERIALSANDMETHODS f (Immunohistological staining) Using the rabbit anti human HGF serum, the sections were immunoperoxidase stained with an avidine biotin technique and nuclei were stained with methvl areen. (In situ hybridization) Mlu I -SalT (O.i’kb) fraament of HGF R chain cDNAwas incerted p Bluescript. -Digoxigenin-labelled se& and antisense HGF RNAswere us~~E;;L;S~be. HGF was detected in the stromal cells, Hofbouer cells and in the basal membrane area where the cytotrophoblast was in contact with the mesenchymal cells by immunohistological staining. By in situ hybridization, HGF was detected in the stromal cells and Hofbauer cells but not in the basal membrane area. [CONCLUSION] HCF was produc ed in mesenchymal cells in placental tissues, and acted on cytotrophoblasts growth factor by paracrine system.
