After sufficient time to insure quantitative coupling and removal of excess reagent, the Nthioacyl polypeptide is subjected to cleavage by acid which affords a 2-substituted-5(4H) thiazolinone of the N-terminal amino acid. Subsequent addition of a large excess of an aliphatic primary or secondary alcohol, either directly to the cleavage acid or after its removal, yields the corresponding N-thioacyl amino acid ester, a stable compound suitable for chromatographic separation and subsequent detection by contemporary methods of high pressure liquid chromatography.

The antibodies are also useful in immunoassays to determine inhibin levels in mammalian fluids. To generate the antibodies, short chain peptides are synthesized which mimic portions of natural mammalian inhibin protein chains, the synthetic peptides are conjugated to large carrier molecules and the conjugates are used as inoculum to induce a mammalian immune system to produce inhibin-reactive antibodies.

An antibody-based bioassay for enzymeinducing chemicals is disclosed. The percent induction of an enzyme or class of related enzymes in living cells by an enzyme-inducing chemical in a sample is determined before and after the addition to the sample of an antibody to the chemical. The concentration of the chemical before and after the addition of the antibody is determined by comparison to the percent induction by known concentrations of the chemical. This permits the determination of the concentration of the chemical in the sample.

The operation is conducted in two stages. The first stage is conducted in the absence of ultrafiltration while the pH of the growth medium is maintained within the range of 6-7 by the addition of a neutralizing agent, Nutrient substratum and dilution water are added during the first stage to maintain the growth rate at a constant level, and the volume of growth medium is permitted to increase. When the amount of growth inhibiting agent produced in the fermentation reaches a predetermined maximum level, the second stage is initiated and the growth rate is maintained in a range of 0.10 to 0.50/hr, which is less than the growth rate in the first stage. By increasing the amount of dilution water the concentration of the growth inhibiting agent is maintained so as to achieve the desired moderate growth rate. The growth inhibiting agent is also continuously removed from the growth medium duringthe second stage and the volume of the growth medium is maintained substantially constant. The extractant employed is chosen from the following groups: (A) double bond unsaturated aliphatic alcohols having 12 or more carbon atoms; (B) saturated branched chain aliphatic alcohols having 14 or more carbon atoms or mixtures thereof; (C) double bond unsaturated aliphatic acids having 12 or more carbon atoms; (D) aliphatic and aromatic mono, di- or tri-esters having 12 or more carbon atoms, other than dibutyl phthalate; (E) aliphatic noncyclic ketones and aliphatic aldehydes having 12 or more carbon atoms; and (F) mixtures of extractants from groups (A) to (E) above or mixtures of at least one of the above extractants and at least one other extractant. These extractants are non-toxic to microorganisms used for fermentation, have little tendency to form emulsions and are otherwise suited for the process. The liquid-liquid extraction may thus be carried out in situ in the fermentor, preferably in a continuous stirred tank fermentor.
