242 4792601

MONOCLONAL ANTIBODY TO MULLERIAN INHIBITING SUBSTANCE Patricia Donahoe, Gerald P Budzik, Meredith Mudgett-Hunter assigned to The General Hospital Corporation In a method for preparing a monoclonal hybridoma which secretes antibodies against an immunogenic material, comprising sensitizing an appropriate source with an immunizing amount of a preparation of the immunogenic material, obtaining from the source lymphocytes having immune activity against the immunogenic material, and fusing the lymphocytes with an appropriate cell line to thereby form mixed hybridomas, the improvement wherein: the immunogenic material is present in the immunizing preparation in an amount not larger than 10% by weight, and wherein, after formation of the mixed hydridomas but prior to cloning, the method comprises the steps of: raising ascites fluid with the mixed hydridomas, then detecting immune specificity against the immunogenic material in the ascites raised fluid; and thereafter cloning the ascites raised cells which show immune specificity against the material.

4792602 ADAPTORS, AND SYNTHESIS AND CLONING OF PROINSULIN GENES Saran A Narang, Ray J Wu, Ottawa, NY, Canada assigned to Cornell Research Foundation Inc A human-like proinsulin gene and its analogs, have been synthesized by a combination of chemical and enzymatic methods. A number o f different human-like proinsulin gene analogs with altered C-chains have also been designed and can be readily constructed as described. As a part of the strategy, an adaptor for trimming DNA has been designed, synthesized and used to recover the A-chain insulin gene with the desired sequence from a hybrid plasmid; a related adaptor for trimming DNA has been designed to shorten the C-chain gene or any gene. The synthetic proinsulin gene has been joined to a replicable cloning vehicle and the hybrid DNA transferred to a host cell. The transformed host

cell has been shown to contain the desired human-like proinsulin gene. 4795700 NUCLEIC ACID PROBES AND METHODS OF USING SAME Peter B Dervan, Geoffrey B Dreyer assigned to California Institute of Technology A probe and method for specific recognition or cleavage of single-stranded DNA or RNA at desired loci utilizing sequence-specific polynucleotide-chelator probes. The probe may also be utilized as a diagnostic agent when the metal ion is replaced with a radiolabelled, fluorescing, or otherwise detectable metal. 4795701 HOMOGENEOUS POLYNUCLEOTIDE DISPLACEMENT ASSAY METHOD KIT AND REAGENT COMPLEX Calvin P H Vary assigned to Allied Corporation An RNA signal strand is displaced from a probe strand by a target nucleotide sequence. Without separation, a digestion enzyme selective for displaced RNA, e.g., a polynucleotide phosphorylase, digests the displaced RNA to nucleoside phosphates including ADP. The digestion products are detected, e.g. by phosphorylating the ADP to ATP and detecting the ATP by biolumineseence.

47~8~ ADHESIN ANTIGENS, ANTIBODIES AND DNA FRAGMENT ENCODING THE ANTIGEN, METHODS AND MEANS FOR DIAGNOSIS AND IMMUNIZATION ETC. Frederick C Lindberg, Bjorn O Lund, Britt M Baga, Mari E Norgren, Mikael Goransson, Bernt E Uhlin, Jan S Normark, David L Lark, Sandviken, Sweden assigned to Syn-Tek AB An antigen which, as its major immunizing component, comprises a determinant of an adhesin
